Could Russia leave US astronauts stranded in space?

A leaky Soyuz decent capsule means that astronauts aboard the International Space Station will have to wait another six weeks before returning home. And private resupply missions are not ready for prime time.

A botched pressure test of a Russian space capsule slated to launch the next crew to the International Space Station has forced NASA and its partners to delay the planned liftoff for more than a month.

The Russian Soyuz spacecraft launch was originally slated for March 29, but now is targeted for no earlier than May 15, NASA's International Space Station program manager?Mike Suffredini told reporters today (Feb. 2).

The Soyuz's crew capsule, one of three modules that make up the entire Soyuz TMA-04M vehicle, ?has been scrapped after an accident during testing caused it to spring a leak in one of its descent module's rocket thruster fuel tanks. Now Russia's main space contractor, RSC Energia, is readying the next spaceship on the line, though the issue will cause a lengthy delay.

"This particular event is very unfortunate, but you know this is a complicated business and things happen," Suffredini said. "To me this is not indicative of some overarching problem at the Energia corporation. With the retirement of NASA's space shuttle fleet last year, Russia's three-person Soyuz space capsules are currently the only vehicle available to ferry American astronauts to and from the International Space Station. NASA plans to eventually use private U.S.-built commercial spacecraft to transport crews and cargo to the station, but those new vehicles are still years away from being ready.

Meanwhile, NASA is contemplating another delay ? in this case, of the unmanned SpaceX Dragon capsule's inaugural trip to deliver supplies to the space station. The private spacecraft was due to make its first test launch to the orbiting laboratory on Feb. 7, but Suffredini said the latest target date is March 20, and even that might be a stretch, as the vehicle still requires more testing and some minor fixes.

The faulty Soyuz capsule was slated to carry NASA astronaut Joe Acaba and Russian cosmonauts Gennady Padalka and Sergei Revin up to the space station to begin long-duration stays.

They were due to replace three of the Expedition 30 spaceflyers currently living on the outpost: NASA astronaut Dan Burbank?and Russian cosmonauts Anton Shkaplerov and Anatoly Ivanishin.

However, because of the Soyuz issues, NASA and Russia have decided to extend the current crew's mission to keep the space station running smoothly. Those three were scheduled to return to Earth March 16, but now will come home April 30.

The delay is also likely to cause rippling setbacks in other planned launches and landings later this year.

Second to Apple, in both profit and revenue, is Samsung, at 16 percent of profit and 25 percent of global revenue. Good numbers, but not nearly Apple numbers.

EDITOR'S PICK: The heart can make 'bad' fat burn calories

Brown adipose tissue (often known as brown fat) is a specialized tissue that burns calories to generate body heat in rodents and newborn humans. Recently, adult humans have also been found to possess brown fat. This piqued the interest of researchers seeking to combat the obesity epidemic, the thought being that increasing the amount of brown fat a person has will make them lose weight. A team of researchers led by Sheila Collins, at the Sanford-Burnham Medical Research Institute, Orlando, has now determined that hormones produced by the heart known as cardiac natriuretic peptides can cause regular fat cells (white adipocytes) from mice and humans to take on characteristics of brown fat cells in vitro. Moreover, infusing one cardiac natriuretic peptide into mice caused regular white fat to upregulate markers of brown fat and increase energy usage. Collins and colleagues hope that with further mechanistic studies it might be possible to exploit these findings clinically to help combat the obesity epidemic.

In an accompanying commentary, Andrew Whittle and Antonio Vidal-Puig, at the University of Cambridge, United Kingdom, highlight the importance of the data generated by Collins and colleagues and discuss the clinical implications of the results.

IMMUNOLOGY: How a stomach-colonizing bacterium protects against asthma

The bacterium Helicobacter pylori can be found colonizing the stomach lining of almost half the world's population. Although persistent infection with Helicobacter pylori increases an individual's risk of developing stomach cancer, it also decreases their risk of developing asthma. A team of researchers led by Anne Mller, at the University of Zrich, Switzerland, has now identified a cellular mechanism by which persistent infection with Helicobacter pylori protects mice from developing allergic asthma. Specifically, they found that Helicobacter pylori modulated immune cells known as dendritic cells such that they did not activate an aggressive immune response but instead activated what is known as a tolerogenic immune response. Although this tolerogenic immune response enabled Helicobacter pylori to persist, it also prevented the onset of unwanted immune responses to allergens (that is, it protected against allergic asthma).

As noted by Kouji Matsushima and Shigenori Nagai, in an accompanying commentary, these data provide new mechanistic insight into the intriguing link between the recent sharp rise in the industrialized world in the number of people with asthma and the simultaneous decrease in exposure to microbes, including Helicobacter pylori.

ONCOLOGY: A sticky problem for stomach cancer

Gastric cancer cancer that arises in the stomach is a leading cause of cancer-related death. A team of researchers led by Jun Nakayama, at Shinshu University Graduate School of Medicine, Japan, has now found that gel-forming mucin secreted by glands in the stomach lining suppresses tumor-promoting inflammation in mice.

Mucin secreted by glands in the stomach lining is characterized by the presence of specific molecules at its ends. These molecules are known as alpha-1,4-linked N-acetylglucosamine residues (alpha-GlcNAc). Nakayama and colleagues found that mice lacking the ability to add alpha-GlcNAc to the ends of mucin secreted by glands in the stomach lining developed stomach cancer. Moreover, expression of alpha-GlcNAc was decreased in the majority of human gastric cancer samples examined. As their previous studies have suggested that alpha-GlcNAccontaining mucin inhibits infection with the bacterium Helicobacter pylori, which is a major risk factor for developing gastric cancer, Nakayama and colleagues suggest that alpha-GlcNAccontaining mucin prevents gastric cancer in two ways: by inhibiting Helicobacter pylori infection and by suppressing tumor-promoting inflammation.

The proteins c-Jun and c-Myc have a key role in promoting cell proliferation, and many human cancers are characterized by aberrant expression of these proteins. Understanding how expression of c-Jun and c-Myc is controlled could provide new insight into the mechanisms underlying tumor formation as well as provide new candidate therapeutic targets. In this context, Kiyotsugu Yoshida and colleagues, at Tokyo Medical and Dental University, Japan, have determined that the protein DYRK2 has a key role in regulating levels of c-Jun and c-Myc in human cells knocking down levels of DYRK2 in human cancer cells increased cell proliferation in vitro and upon transplantation into mice. Moreover, expression of DYRK2 was decreased in tissue samples of many different types of cancer, including breast, colon, prostate, and kidney cancers. Yoshida and colleagues therefore suggest that restoring DYRK2 expression could provide a new approach to treating many different forms of cancer.

ONCOLOGY: Promoting instability in cancer cells

Many cancer cells are characterized by gain or loss of whole chromosomes and/or major structural changes to chromosomes. These chromosomal changes, which are said to indicate chromosomal instability, often drive tumor formation and/or progression by causing overexpression of tumor-promoting proteins or generation of tumor-promoting fusion proteins. Understanding the mechanisms underlying chromosomal instability should provide new insight into the mechanisms underlying tumor formation/progression as well as provide new candidate therapeutic targets. In this context, a team of researchers led by Richard Pestell, at Thomas Jefferson University, Philadelphia, has now determined that the protein cyclin D1 induces chromosomal instability and increased breast tumor prevalence in mice. Moreover, analysis of human breast tumor samples indicated that cyclin D1 expression correlated with chromosomal instability in the luminal B subtype of breast cancer. Additional experiments indicated that cyclin D1 mediated its effects by increasing expression of numerous genes related to chromosomal instability. Further analysis of this mechanism could provide new targets for anticancer therapies.

Gastric cancer cancer that arises in the stomach is a leading cause of cancer-related death. A team of researchers led by Jun Nakayama, at Shinshu University Graduate School of Medicine, Japan, has now found that gel-forming mucin secreted by glands in the stomach lining suppresses tumor-promoting inflammation in mice.

Mucin secreted by glands in the stomach lining is characterized by the presence of specific molecules at its ends. These molecules are known as alpha-1,4-linked N-acetylglucosamine residues (alpha-GlcNAc). Nakayama and colleagues found that mice lacking the ability to add alpha-GlcNAc to the ends of mucin secreted by glands in the stomach lining developed stomach cancer. Moreover, expression of alpha-GlcNAc was decreased in the majority of human gastric cancer samples examined. As their previous studies have suggested that alpha-GlcNAccontaining mucin inhibits infection with the bacterium Helicobacter pylori, which is a major risk factor for developing gastric cancer, Nakayama and colleagues suggest that alpha-GlcNAccontaining mucin prevents gastric cancer in two ways: by inhibiting Helicobacter pylori infection and by suppressing tumor-promoting inflammation.

One of the hallmarks of cancer is the ability of cancer cells to proliferate in an uncontrolled manner. The proteins c-Jun and c-Myc have a key role in promoting cell proliferation, and many human cancers are characterized by aberrant expression of these proteins. Understanding how expression of c-Jun and c-Myc is controlled could provide new insight into the mechanisms underlying tumor formation as well as provide new candidate therapeutic targets. In this context, Kiyotsugu Yoshida and colleagues, at Tokyo Medical and Dental University, Japan, have determined that the protein DYRK2 has a key role in regulating levels of c-Jun and c-Myc in human cells knocking down levels of DYRK2 in human cancer cells increased cell proliferation in vitro and upon transplantation into mice. Moreover, expression of DYRK2 was decreased in tissue samples of many different types of cancer, including breast, colon, prostate, and kidney cancers. Yoshida and colleagues therefore suggest that restoring DYRK2 expression could provide a new approach to treating many different forms of cancer.

Many cancer cells are characterized by gain or loss of whole chromosomes and/or major structural changes to chromosomes. These chromosomal changes, which are said to indicate chromosomal instability, often drive tumor formation and/or progression by causing overexpression of tumor-promoting proteins or generation of tumor-promoting fusion proteins. Understanding the mechanisms underlying chromosomal instability should provide new insight into the mechanisms underlying tumor formation/progression as well as provide new candidate therapeutic targets. In this context, a team of researchers led by Richard Pestell, at Thomas Jefferson University, Philadelphia, has now determined that the protein cyclin D1 induces chromosomal instability and increased breast tumor prevalence in mice. Moreover, analysis of human breast tumor samples indicated that cyclin D1 expression correlated with chromosomal instability in the luminal B subtype of breast cancer. Additional experiments indicated that cyclin D1 mediated its effects by increasing expression of numerous genes related to chromosomal instability. Further analysis of this mechanism could provide new targets for anticancer therapies.

Lee Adler, historic preservationist, dies at 88 (AP)

SAVANNAH, Ga. ? Lee Adler II was never content with saving Savannah's historic homes and buildings one at a time. In 1959, he found a way to spare entire city blocks and neighborhoods from the wrecking ball that changed the way preservation groups did business ? not just in Georgia, but across the nation.

Adler died Sunday at age 88, said Matt Weeks of Fox & Weeks Funeral Directors. A cause of death was not immediately available.

As the president of the Historic Savannah Foundation, which Adler led until the mid-1960s, he took an entrepreneurial approach to saving Savannah's architectural treasures by essentially persuading local preservationists to get into the real estate business. The group would buy sagging old properties facing demolition and sell them to buyers who promised to restore them. The tactic worked so well in Savannah that groups across the U.S. began following Adler's lead.

"There's not a preservation group in this country that doesn't owe some debt of gratitude to the work of Lee Adler," said David J. Brown, chief preservation officer at the National Trust for Historic Preservation in Washington.

Adler, usually working in tandem with his wife, Emma, became an evangelist for the methods he'd used in Savannah. He would tout his hometown's success at preservation conferences and conventions and even to individual groups across the nation. He also co-wrote a handbook on the subject in 1974.

"Lee's the kind of person who was a lot more respected outside of Savannah than he was in Savannah, because inside the city a lot of people took preservation for granted," said Mark McDonald, a longtime friend of Adler and the president of the Atlanta-based Georgia Trust for Historic Preservation. "He gave people the idea they could do this in their hometown just like they'd done it in Savannah."

The influence was noted in "Midnight in the Garden of Good and Evil," John Berendt's best-selling book about a murder in Savannah. Berendt wrote that because of Adler's work in historic preservation, "Lee Adler was probably the best-known Savannahian outside Savannah" by the 1980s.

Born into a wealthy Savannah family that owned a local department store, Adler's passion for protecting the 18th and 19th century homes of Georgia's oldest city was passed on by his mother.

Elinor Grunsfeld Adler was among the seven women who launched the foundation that her son would later lead. The women started the group in 1954 to show their outrage after the downtown City Market, where farmers sold their crops, was razed to make way for a parking garage.

The preservation group saved other homes and buildings, typically turning them into museums, during a time when the suburban flight that followed World War II left Savannah's oldest neighborhoods decaying and abandoned. The fledgling foundation's methods would change dramatically under Lee Adler's leadership years later.

In 1959, he learned a wrecking company had a permit to demolish four century-old townhouses in downtown Savannah with the intention of selling the bricks for a profit. Adler quickly struck a deal to buy the entire row for $54,000 and got the Historic Savannah Foundation's members to agree to share the cost ? $180 for each of its 300 members. The homes were later sold to new owners.

Daniel Carey, the foundation's current president and a longtime friend of Adler, said the revolving-fund technique Adler popularized is credited with saving more than 350 homes and buildings in Savannah, where the 2.2-square-mile downtown area forms America's largest National Historic Landmark District.

"Historically there were efforts to save one building at a time, call it a success and kind of take a victory lap and be finished," Carey said. "Lee and others said this is really just the beginning, not the end. So they said we can save a block at a time and neighborhood at a time ? and eventually a city at a time."
